Molecular characterisation of a ‘Candidatus Phytoplasma australasiae’ strain associated with Jasminum sambac phyllody in India

Abstract

Symptoms of yellowing and little leaf were observed in jasmine plants at farmer fields of Kancharapalem village, Varadaiahpalem Mandal, Tirupati, India. DNA was extracted from symptomatic and asymptomatic leaves and subjected to nested PCR assay with P1/P7 and R16F2n/R16R2 primers and secAfor1/sArev3 and SecAfor2/SecArev3 primers. The PCR amplified 1,250 bp and 480 bp products were eluted, sequenced and the aligned sequences obtained were submitted to NCBI GenBank. Sequence analysis of 16Sr RNA and SecA genes through phylogeny and virtual RFLP of jasmine phyllody phytoplasma allows its identification as ‘Candidatus Phytoplasma australasiae’ and its assignment to 16SrII-D subgroup, respectively.
