Potential use of enzyme conjugated Anti-Carp Immunoglobulin in detecting specific antibodies in carp

Enzyme immunoconjugates are one of the key components of ELISA which so far, commercially not available in case of fish. In this study an attempt was made to develop anti-carp immunoglobulin enzyme conjugate and to assess its potential in carp disease diagnosis. Immunoglobulins (Ig) were isolated from the Indian Major Carps (IMC) and hyperimmune serum was raised in rabbit against carp Ig. In immunodiffusion (ID) and countercurrent immunoelectrophoresis (CIE), specificity between carp Ig and rabbit anti-carp Ig was observed. Anti-carp Ig enzyme conjugate was prepared by coupling horseradish peroxidase (HRPO) to rabbit anti-carp Ig. The immunoconjugate prepared showed reactivity to carp Ig and also cross reacted to sera from rohu, catla, mrigal and common carp through direct ELISA. The immunoconjugate was also used to detect the antibody response of Aeromonas hydrophila sensitized laboratory maintained carp through indirect ELISA, indicating potentiality of the conjugate in detecting different anti-pathogen antibodies in carp sera.
