Effect of biopriming on ajwain (Trachyspermum ammi L.) seed germination and vigour

Abstract

A laboratory study was conducted to evaluate the efficacy of different biopriming agents in combination with chitosan solution for the management of seed germination and poor seedling growth of ajwain (Trachyspermum ammi L.). The surface sterilized seeds of ajwain were soaked in 0.2 per cent chitosan solution (made in 1.0 per cent acetic acid) for 3 hours. The pH of chitosan solutions was adjusted to 6.0 with help of 1.0 % sodium hydroxide solution. After that, a freshly grown culture broth of plant growth promoting rhizobacterial isolates or Trichoderma sp. were applied on these pre-soaked seed. 0.2 ml per gram culture broth of each rhizobacterial isolates or Trichoderma sp. (5.0×107 cfu mL–1) were applied on chitosan pre-soaked seeds and were mixed thoroughly under asceptic conditions. The highest germination (94.44 per cent) was recorded with chitosan + Pseudomonas fluorescens + Pseudomonas aeruginosa which was at par with Chitosan alone, followed by Chitosan + Bacillus megaterium + Paenebacillus polymyxa. The lowest seed germination (74.67 per cent) was recorded with control. The seed germination was significantly hastened by the biopriming treatments irrespective of combination of bio-agents in comparison to control. The different bio priming agents showed significant effect on rate of seed germination in ajwain. The highest rate of seed germination (6.63) was recorded with Chitosan + Pseudomonas fluorescens + Pseudomonas aeruginosa followed by Chitosan + B. megaterium + B. subtilis (6.53) and lowest was recorded with control (4.77). This is the first report of combined application of PGPR and chitosan for enhancing ajwain seed germination in India.
