Development of an Efficient Micropropagation System in Banana cv. Malbhog (AAB)

Abstract

Thebanana cultivar Malbhog withthe most appetizing and high valued fruit in India particularly Bihar is facing extinction due to Panama wilt disease. An efficient micropropagation protocol was developed culturing shoot apices from sword suckers on MS, B5 and modified MS media with phytohormones. The best medium for caulogenesis was M14 (B5 + 1.14 μM IAA+ 19.97 μM BAP) with the maximum frequency (94.79%) and the highest number of differentiated shoots per culture (24.75). Another medium M11 (MS+1.14μM IAA+19.97 μM BAP) was found at par for caulogenesis. The subculture of differentiated multiple shoots on medium M11 resulted in cent percent shoot multiplication with the number of differentiated shoots per culture remaining constant upto fifth subculture and thereafter declined-3.37% in sixth to-40.45% in eight subculture. The best medium for rhizogenesis was M17 (MS+4.92 μM IBA). The banana plantlets were progressively acclimatized and successfully transferred to field conditions.
