Phytochemical investigation, chemical composition and in vitro antioxidant activities of various crude extracts of Holigarna ferrugenia Marchand

Abstract

Plants are considered to be the repositories of various phytoconstituents that are medicinally important to mankind. In the present study, preliminary phytochemical analysis, quantification of total phenolics (TPC), total flavonoids content (TFC) and assessment of antioxidant potential of Holigarna ferrugenia Marchand (HF) leaves were investigated. The dried powdered leaves of H. ferrugenia were subjected to Soxhlet extraction using solvents viz., hexane, ethyl acetate and methanol. The crude extracts obtained were further analysed using GC-MS to detect the secondary metabolites present in the extract. Colorimetric methods was used to quantify TPC and TFC from the crude extracts of HF leaves. Furthermore, DPPH assay, FRAP assay, and phosphomolybdenum (PM) assay were exploited to determine free radical scavenging activities. Phytochemical analysis of various solvent extracts showed presence of major classes of phytochemicals. Also, the GC-MS reports revealed the presence of 13 active compounds in hexane extract, 9 in ethyl acetate and 5 compounds in methanolic extract. Ethyl acetate extract was found to contain the highest TPC and TFC. In vitro free radical scavenging activities of crude extracts showed highly significant result which are comparable with the standard ascorbic acid. Our results suggest that the crude extracts of H. ferrugenia contains rich source of polyphenols and antioxidants that can play an important role in preventing the progression of various health ailments.
